Instrument Specification

Specifications of the Rigaku OD (Oxford Diffraction) XtaLAB Synergy-DW Custom X-ray diffractometer:

  • Rigaku MicroMax-007HF DW Generator – microfocus rotating anode X-ray generator with dual-wavelength capability (Mo Kα or Cu Kα)

  • Rigaku VariMax DW Optics – dual-wavelength confocal X-ray optics

  • HyPix-6000HE – hybrid photon-counting (HPC) X-ray detector 

  • UG2 – second-generation universal 4-circle κ-goniometer for XtaLAB Synergy diffractometers 

  • Oxford Cryosystems Cryostream 700 – open-flow cryogenic cooling system

  • Conventional goniometer head with magnetic base and LithoLoopsTM  mounted in CryoCapsTM  as crystal holders

  • Rigaku Oxford Diffraction CrysAlisPro  - diffractometer control and data acquisition software

Sample Preparation and Transport

The purpose of sample preparation is not to assess crystal quality in advance or to select only the best crystals, but to prevent changes or degradation of potentially suitable single crystals during transport.

Basic recommendations:

  • Store samples in closed containers (e.g. microtubes, vials, NMR tubes) to prevent drying or solvent loss

  • Use transparent containers to allow visual inspection of the sample under a stereomicroscope

  • Choose containers that allow easy handling; avoid vials with narrow or screw-top necks

  • Minimize exposure to air for samples that may be unstable

Types of Analyses and Experiments

  • SM single-crystal diffraction: Single-crystal X-ray diffraction experiment performed on the small-molecule (SM) sample, aimed at determining the unknown 3D structure of the sample.

  • (p)XRD experiment: X-ray diffraction experiment performed on powder or polycrystalline samples (i.e. non-single-crystal samples).

  • SM finalization: Refinement and completion of a preliminary CIF file into a form suitable, for example, as supplementary information for a scientific publication. 

Requests for PX diffraction experiments (= X-ray diffraction on protein single crystals) should be submitted via the CF BIC booking system.
